They were not biological brothers. They were brothers in the sense that all firefighters are brothers or police officers are brothers. They were both supposed to be concerned with the welfare of the Matoran.

I have actually never been told who the villain behind the brains is. At one point, I had suggested doing a book on their origins, but I was told not to, that there were already plans to reveal the power behind them at some point. So I don't have info to answer you, sorry.
